چکیده

Epithelial sodium channels (ENaCs) controls vectorial Na across epithelia thus playing a central role in all aspects of fluid clearance in the body. The aim of this study was to investigate in vitro time dependant model of hypoxia induced reduction in vectorial Na and fluid transport across the alveolar epithelium. AEC exposed to 3% O2, for different time periods (1 h, 3h, 6h, 12h, 24h and 48 h) showed increased oxidative stress (↑ROS,↑MDA) and NF-κB up-regulation. Hypoxia induced a time-dependent decrease in Na/KATPase activity and α1 Na /K-ATPase protein expression reaching to negligible levels after 6h of exposure. Intriguingly, hypoxia also decreased copy number of ENaC α-subunit in AEC beyond detectable levels after 12h. Curcumin reinstate survival signalling by blunting NFkB signalling and reinforced the levels of alveolar antioxidants along with significant (p<0.001) increase in ENaC and Na/K-ATPase expression in lungs of rats under hypoxia. Therefore, we propose that, curcumin prophylaxis abate NFkB aberrant signalling induced down regulation of Na/K-ATPase and ENaC under hypoxia.

Diabetic Encephalopathy Affects Mitochondria and Axonal Transport Proteins

Introduction: Diabetic encephalopathy is described as any cognitive and memory impairments and associated with hippocampal degenerative changes, include neurodegenerative process and decreased number of living cell. Mitochondrial Diabetes (MD) appears fallowing activation of mutant mitochondrial DNA and is combination of diabetes and cognitive deficit. In this research we showed the correlation...

Hypoxia upregulates activity and expression of the glucose transporter GLUT1 in alveolar epithelial cells.

Alveolar epithelial cells (AEC) are directly exposed to high alveolar O(2) tension. Many pulmonary disorders are associated with a decrease in alveolar O(2) tension and AEC need to develop adaptative mechanisms to cope with O(2) deprivation. Under hypoxia, because of inhibition of oxidative phosphorylation, adenosine triphosphate supply is dependent on the ability of cells to increase anaerobic...

Dexamethasone prevents transport inhibition by hypoxia in rat lung and alveolar epithelial cells by stimulating activity and expression of Na+-K+-ATPase and epithelial Na+ channels.

Hypoxia inhibits Na and lung fluid reabsorption, which contributes to the formation of pulmonary edema. We tested whether dexamethasone prevents hypoxia-induced inhibition of reabsorption by stimulation of alveolar Na transport. Fluid reabsorption, transport activity, and expression of Na transporters were measured in hypoxia-exposed rats and in primary alveolar type II (ATII) cells. Rats were ...

Modulation of cellular transport characteristics of the human lung alveolar epithelia

Among the drug delivery and targeting (DDT) routes, lung alveolar epithelium has been given enormous attentions in terms of the delivery of a wide range of macromolecules such as gene- or protein-based nanopharmaceuticals. However, little is known about cellular modulation of lung transport characteristics by endogenous and/or exogenous agents. Thus, in the current study, impact of dexamethason...

Hypoxia reversibly inhibits epithelial sodium transport but does not inhibit lung ENaC or Na-K-ATPase expression.

Hypoxia reduces alveolar liquid clearance and the nasal potential difference, a marker of airway epithelial sodium transport. The mechanisms underlying this impaired epithelial sodium transport in vivo remain uncertain. We hypothesized that epithelial sodium transport impaired by hypoxia would recover quickly with reoxygenation and that hypoxia decreases the expression of lung epithelial sodium...
